Modifications to backbonePlasmid assembled from 3 CYANO-VECTOR modular devices: pCVD047 / CV-RSF1010Y25F: The device carries a broad host range replicon derived from RSF1010. A point mutation in mobA prevent self-mobilization. pCVD002 / CV-aadA: The device confers spectinomycin and streptomycin (SpSm) resistance. The stop codon was erased by mistake, another stop codon is however located 15 nt downstream in the transcriptional terminator. This device works perfectly well. pCVD030 / CV-PconII-LTRBS-GFPmut2: Promoter/reporter device where GFPmut2 is driven by a PconII promoter. The RBS and flanking sequences are from the pENTR/SD/D-Topo vector (Life Technologies).
