Modifications to backboneA scaffold with interspersed restriction sites containing genes encoding chloramphenicol and gentamicin resistance and GFP was inserted into the backbone. In addition, the temperature-sensitive origin of replication ori101/repA101ts was inserted between the blunt restriction sites PsiI and SnaBI, destroying the R6K origin of replication. A synthetic kill switch consisting of a gene encoding the Tet repressor and a tet-controlled synthetic operon containing the genes hokB, ghoT, and tisB was also inserted into the backbone.
