Backbone

• Vector backbone: pUC18R6K
• Modifications to backbone: A scaffold with interspersed restriction sites containing genes encoding chloramphenicol and gentamicin resistance and GFP was inserted into the backbone. In addition, the temperature-sensitive origin of replication ori101/repA101ts was inserted between the blunt restriction sites PsiI and SnaBI, destroying the R6K origin of replication.
