Depositor Comments

Plasmid containing K-ras G12D with a LoxP-STOP-LoxP cassette included upstream of the K-ras sequence such that K-ras G12D will only be expressed after cre-mediated recombination.

The positive selection for ES cells is puromycin, and negative selection is carried out by addition of ganciclovir.

The plasmid sequence submitted by the author is an approximation. For example, the Bluescript backbone sequence was chosen randomly (e.g. the Sal I site in the backbone is definitely not present in the construct).
