Mouse IRS-4 genomic DNA was screened from the BAC genomic DNA library (Genome Systems Inc. St. Louis, Mo.) by PCR with specific primers for mouse IRS-4 genomic DNA. The AvrII-ClaI fragment encompassing the entire exon 1 of mouse IRS-4 gene was blunt ended and ligated into the SnaBI site in pBABE-puro.
