We derived the tAD-seq Gal4-DBD-candidate expression plasmid from the fly STARR-seq plasmid (AddgeneID 71499) (Arnold et al, 2013) by replacing sgGFP with the Gal4-DNA-binding domain (DBD) followed by a poly-glycine linker upstream of the candidate library insertion site, containing the ccdB suicide gene flanked by homology arms (used for cloning the candidates during library generation; for the library cloning strategy please see ref. (Arnold et al, 2013)), which is followed by three stop codons (one for each reading-frame).
