This gel depicts the NEB 1kb ladder and a digestion of the plasmid with the following enzyme: HindIII
We expected the following band sizes for this digest: 3.4kb and 6.6kb. We did not see the expected pattern--the plasmid was only linearized (though the linearized plasmid was the correct size). Since we see the HindIII site at the 5' end, we have changed the data to reflect the destroyed 3' HindIII cloning site.
