TIMER-bac was generated by exchanging TCC coding for serine 197 to ACC coding for threonine in DsRed.T3_S4T (Sörensen et al., 2003). TIMER-bac was cloned downstream of the constitutively active PybaJ promoter (Rollenhagen and Bumann, 2006) on a pBR322-based plasmid carrying a bla gene conferring resistance to ampicillin (“pBR322_TIMER”).
