  • Purpose

    This pair of duplicate libraries contains SARS-CoV-2 spike receptor binding domain (RBD) mutants in a yeast surface display vector.

    Each library is barcoded with randomized N16 barcodes downstream of the coding sequence.

    These libraries can be used for comprehensive mapping of the biochemical effects of RBD mutations.

Library Shipping

This library will be delivered as two pooled DNA libraries in microcentrifuge tubes on blue ice (tubes labeled lib1 and lib2). The tube contents will not necessarily be frozen. For best results, minimize freeze/thaws.

For each library tube, you will receive:

A scheme illustrating library assembly and use. In step one, a cartoon representation of PCR-based codon mutagenesis showing DNA with NNS sites leading to a library of mutant variants. In step three, random barcode sequence is appended to each mutant gene, and PacBio sequencing is used to generate long reads spanning barcode and mutant gene to generate a barcode-variant lookup table. The barcoded gene library is then expressed in a yeast display system, and Illumina sequencing of short barcodes can be used to read out the mutant variant in experimental partitions.
