Depositing Lab: Ian Frew
The Multiple Lentiviral Expression (MuLE) is a modular and flexible system that can be used to construct complex lentiviruses for modification of mammalian cells with a single viral infection.
This kit contains 96 plasmids and is shipped as bacterial glycerol stocks in a 96-well plate format.

Description
The MuLE (Multiple Lentiviral Expression) system facilitates the simultaneous introduction of multiple genetic alterations into mammalian cells. The MuLE system kit consists of 91 building block Entry vectors and 5 lentiviral Destination vectors to allow simple and rapid MultiSite Gateway recombination-based cloning of complex polycistronic lentiviruses containing up to five different genetic expression elements. These vectors can induce combinatorial constitutive or inducible gene overexpression, knockdown (shRNA, miR-30-shRNA), mutation/editing (CRISPR-Cas9) or deletion (Cre), together with expression of fluorescent or enzymatic reporters for cellular assays and animal imaging studies. The ability to simultaneously alter multiple genes via infection with a single ecotropic or amphotropic MuLE lentivirus provides significant genetic power and allows high throughput genetic modulation studies in mammalian cells and tissues.
Overview of MuLE vector assembly
Schematic overview of a recombination reaction involving four MuLE Entry vectors and a lentiviral Destination vector, allowing combinatorial expression of combinations of the listed genetic elements. A series of vectors with similar genetic elements are also available to allow recombination reactions involving two or three Entry vectors
