Modifications to backbonepcDNA6.V5.HisB vector was modified with NheI and PmeI restriction digestion enzymes to cut out MCS, V5 and His tag sites. New multiple cloning sites using annealed primer pair were ligated back into the vector. The newly generated cloning site comprises: NheI, KpnI, EcoRV, XhoI and SacII. Codon optimized sT was cloned between EcoRV and XhoI restriction sites.
